Yes. For one Build Option, you can add as many upgrades as you would like to one of your spires in play - as long as you do not exceed your spire's upgrade capacity.
Note that constructing a spire and upgrading that spire would be two separate Build Options. You cannot construct a spire and immediately upgrade it using the same Option.
